Evaluación de métodos de marcación-radioactivos y no radioactivos empleados en la elaboración de sondas diagnósticas de malaria
Abstract
We report here a comparison of various methods for labelling of Plasmodium falciparum DNA probes. The sensitivity of P. falciparum DNA detection was increased ten times when the random primers method was used instead of the nick translation method. Using the probe PFCOL692 developed in our laboratory, the detection level was 12 pg of DNA. These results are similar to those reported by other laboratories using the probe pRepHind. When compairing non-radioactive methods for labeling, photobiotin was adventageus in comparison with nick translation biotin-dUTP. The detection limit was 135 pg of P. falciparum DNA. A fluorescent system based on proteases and fluorogenic substrates was assayed, and its expectations were discussed.
